LIMITED PROTEOLYSIS OF ALLERGEN ARA H1, STORAGE 7S GLOBULIN FROM PEANUT SEEDS
A hypothetical scenario of papain limited proteolysis of storage 7S globulin Ara h1, the strongest allergen from peanut seeds, is described. The scenario is based on the results of electrophoresis of proteolysis of high-molecular-mass products as well as on the available information on the presence of disordered regions potentially susceptible to proteolytic attack in the Ara h1 subunits. It was demonstrated that the initial Ara h1 limited proteolysis leads to the detachment of the subunit N-terminal disordered region containing one third of the total number of antigen determinants (IgE epitopes) identified in the Ara h1. Obviously, the N-terminal region teeming with arginine residues, which correspond to papain specificity, is completely destroyed during proteolysis. Thus, the level of Ara h1 allergenicity can be decreased via limited proteolysis. Keywords: seed storage 7S globulins, allergens, IgE epitopes, peanut.
